<p>Daerah  fungsional  adalah  daerah  DNA  eukariotik  di  sekitar  gen  yang  dapat<br />
diidentifikasi  dengan  diberi  perlakuaan  DNAse  I,  sedangkan  fungsi  utama  dari  daerah<br />
fungsional adalah untuk mengkode protein supaya gen itu dapat di ekspresikan.</p>

Insulator merupakan daerah yang membatasi daerah funngsional yang berukuran 1-<br />
2 kb. Fungsi utama insulator adalah sebagai berikut:<br />
1.  Menghilangkan  efek posisional,  yaitu  ekpresi gen  yang disisipkan ke dalam  suatu<br />
kromosom eukariot daerah kromatin yang kompak dimana gen tersebut inaktif tanpa<br />
insulator<br />
2.  Menjaga independensi dan mencegah “cross talk” antara domain-domain fungsional<br />
yang berurutan dengan gen yang terikat pada insulator.</p>

<p>Fungsi  insulator  ini  didukung  oleh  protein  pengikat  DNA,  pada  Drosophilla  Su<br />
(Hw)  yang  secara  khusus  terikat  oleh  isulator.  Daerah-daerah  yang  sensitive  terhadap<br />
DNAse  I  terletak  di  sepanjang  kedua  sisi  gen-gen  yang  diekspresikan  dan  memiliki<br />
konfigurasi  terbuka.  Domain  fungsional  ini  dibatasi  oleh  saequense  insulator  scs<br />
(spesialised chromatin structure) yang terdapat pada kedua sisi gen hsp dari lalat buah.</p>
<p>Locus  Control  Region  (LCR)  adalah    suatu  urutan  DNA  yang  berperan  dalam<br />
pembentukan dan pertahanan domain fungsional yang  terbuka dalam konfigurasi aktif dan<br />
juga dapat mengurangi efek posisional, misalnya pada LCR globin yang terletak sekitar 12<br />
kb upstream gen-gen  globin. LCR globin mengandung 3  situs DNA hypersensitive, pada<br />
situs  ini  nukloesom  mengalami  modifikasi  karena  sifatmnya  aksesibel  terhadap  protein<br />
pengikat DNA. Protein tersebut mengontrol struktur kromatin dalam domain fungsional.</p>
